Annihilation introduit une toute nouvelle façon de jouer au jeu. Poussez plus profondément dans la grotte pour atteindre les points de faiblesse structurelle, posez de puissantes bombes, puis revenez à la sortie avant qu'elles n'explosent. L'Exterminateur peut empêcher les horreurs de se reproduire et est immunisé contre les gaz, ce qui vous aide à garder vos voies d'évacuation ouvertes pendant que vous terminez votre mission.
